Acid phosphatase is one of a group of enzymes located primarily in the prostate gland and prostatic secretions. Smaller amounts are found in the bone marrow, spleen, liver, kidneys, and blood components such as erythrocytes and platelets.

An Acid Phosphatase test is presumptive test for semen. It may be performed directly on a cut out portion of a stain, an extract of a stain, or a “wipe” of the stained material. A wipe may be made using a piece of filter paper, thread, or swab.

Total acid phosphatase (AP) activity may be useful when evaluating for prostate cancer, Paget’s disease, hyperparathyroidism with skeletal involvement, and Gaucher’s disease since elevations in AP activity occur in these conditions.
